!! The New Polymer Standards with linear structure and expanded conformation
High performance size exclusion chromatography (HPSEC) is frequently used for the measurement of Mw and molecular weight distribution (MwD). Unless the system is equipped with a light scattering detector, molecular weight standards are needed to calibrate the HPSEC columns. Sometimes even the system is equipped with a light scattering detector, since the accuracy of light scattering is not adequate for measuring Mw below 200,000, Mw standards are yet needed to determine the Mw of such polymers. Pullulan and dextran are commercially available Mw standards, but since the retention time in SEC is dependent on hydrodynamic volume rather than Mw, the use of pullulan and dextran as standards can lead to significant overestimation of Mw of polysaccharides with different backbone and more expanded chain conformation. Besides, both pullulan and dextran are 1-6-linked glucans. The three-bond glycosidic linkage through 1-6 increases the freedom of rotation between two units, as a result, the polymer chains show a very flexible chain conformation in solution. Putus is now providing PS-OBGs, the aqueous polymer standards originated from oat bran. Our research showed that PS-OBG standard possesses a linear chain with mixed linkage of (1?3) and (1?4) and the ratio of tri- and tetra- building blocks is ~2:1. The structural characterization indicated that PS-OBG have a more expanded chain conformation compared with pullulan and dextran. In terms of accurately measuring Mw by SEC, standards of PS-OBG should be more suitable since its structure are more uniform and the hydrodynamic volume (molecular size) is directly related to the type of glycosidic linkages and branching.
!! Larger Range of Mw and Narrower Mw distribution
Putus offers PS-OBG, a new carbohydrate polymer molecular weight (Mw) standards with Mw ranged from 10,000 to 1,400,000 and Mw distribution (MwD) of 1.01 – 1.05. Compared to the currently available polymer standards pulullan and dextran (Mw ranged from 5900 to 788,000), PS-OBG demonstrated a larger Mw range which facilitate the Mw measurement of very large polymers. Pullulan and dextran are universally used as polymer standards because so far there is no commercially available beta-glucans that are comparable with pullulan and dextran with regards to the narrow Mw distribution. The PS-OBG produced by Putus demonstrated an extremely narrow MwD of 1.01-1.05. For the standards with Mw below 200,000, the MwD can reach 1.001-1.005.
